Judging from this I'd say that the second set of speakers you mentioned, that RCA to 3.5mm cable, a DAC external sound card and this amp is all compatible to each other and thus, should work!

Now, the ValveDock has an Audio In in the form of the ordinary Red and White plugs, also known as RCA. The audio L & R on this picture:
Now, to connect this to your computer and bypassing your ordinary sound card, you need a DAC who works as an sound card, like this one: This allows you to add one 3.5 speaker plug and one 3.5mm mic plug. Now, this one allows you to create a sound card which you can connect your amp to. And to connect, you need an 3.5mm to RCA cable to connect these two. Like this:

Yep, the RCA to 3.5mm is a completely analogue cable.

Well, connecting the speakers to the amp, if you mean the cables coloured dark gray and dark red on the image.
